Spectrophotometer Instrument instructions

Spectrophotometer Instrument instructions
Purpose
The purpose of this guideline is to describe the process of work and quality control of the spectrophotometer.
2- Scope of application
This guideline applies to biochemistry and hematology.
3- Responsibility for implementation
Responsible for the implementation of this implementation method with the technical staff of the Department of Biochemistry, Hematology and is responsible for quality assurance.
4- Definitions
does not have
5- Description of actions
5-1- Principles.
Refer to the Reference Laboratory Booklet.
5-2- Method of work:
1- Turn on the Power button on the back of the device (black button)
2- After turning on the device, first a series of tests, including
- System Test
- Dark Calibration
- Wavelenght Calibration
It is done automatically (note that before turning on the device, the sample housing should be checked so that the couture is not placed inside the light path).
3- After this step, the Genova screen will appear, in which the various modes used will appear:
- Photometric
- DNA / RNA Mode
- Protein Mode
- Instument Setup
- Security
4- Go to the Photometric section and press Enter.
5- Using the arrow-like buttons, come to the Setup section and press Enter.
6- Using the arrows, we come to the Wevelenth item (wavelength) and using the arrows, we come to the right side and select the desired wavelength and press Enter.
7- Re-exit the item and press Enter.
8- Open the door of the compartment to put the couture and after washing the couture with distilled water and pouring the distilled water into it (at least 1ml), put it in its place and close the lid and press the CAL button to calibrate the device to zero.
9- Then we pour the solution that we want to measure its light absorption into the couture (after removing the distilled water from the couture, we read its edge and after at least 10 seconds, the ABS level is read.
6- Control and maintenance:
Spectro photometers should be subjected to qualitative control in 6-month periods in terms of linear control (Linearity), photo-metric accuracy, wavelength accuracy, stability test, or photo-meter drift. The above method is attached to the Iran Reference Laboratory Booklet.
The spectrophotometer should be placed in a dry place away from direct sunlight, and after working with it, turn off the device and put the cover on it.
6-1- Quality control solutions:
Solution No. 1 - Dry potassium dichromate powder in Oven at 110 سان C for one hour and 100 mg with 0.01 normal sulfuric acid to a volume of one liter. The solution is stored as a stock in a dark glass. do. This solution is used to control linearity.
Pour 101 ml of 0.01 N normal sulfuric acid into the ten-tube tube, except for the No. 1 tube, and add 2 ml of the stock solution to the No. 1 tube. Mix the No. 2 tube well and transfer 2ml of it to the No. 3 tube, and so on. We go to the pipe number 10. (We prepare the serial dilution.)
Solution No. 2 - Solution in hemoglobin solution (used to control the accuracy of wavelength or λ Max)
Solution No. 3- Dilute one-half of solution number one (be sure to prepare in class A jelly balloon). This solution is used to control photometric accuracy.
Solution No. 4 - Solution in hemoglobin derivatives. To check the control of photometric thrust
((Drift is used.
Then fill the spectrophotometer quality control form.
7- Service and repair:
In case of any disturbance in the control parameters, the support company should be contacted and serviced as soon as possible and its report should be recorded and kept in the service and maintenance form.
8- Safety considerations for working with the device:
1- Do not put the couture inside the OD reading chamber after finishing the work.
2- Do not pour any solution inside the OD reading chamber.
3. Make sure the coots are clean before starting work. (Be clear and transparent)
4- It is necessary to use the current regulator system (transformer) before connecting the spectrophotometer to the city electricity.
5. It should be placed away from sunlight, vibration, acidic or alkaline chemicals.
6- Sample area should always be clean and clean as soon as the sample is contaminated. External surfaces of the device should always be clean and free of dust.
7- When not using the device, it is recommended to pull the two branches from the socket and put a cover on it.
8- If you see a photometric inaccuracy, refer to the brochure and control the specifications of the lamp used to replace or troubleshoot it.
9- Since Spectrophotometer is an accurate and sensitive tool, do not take any action to replace parts without the approval of the relevant agency.
